Molecular Loop’s chemistry comprises only 4 steps, unlike other enrichment chemistries. Molecular inversion probes (MIPs) are designed to target regions of interest in the sample DNA. The MIPs are hybridized to sample DNA and circularized enzymatically, and then unique molecular identifiers (UMIs) are introduced. Unique dual-index adapters are then added to each DNA fragment by PCR to create the sequencer-ready library. These genetic elements enable sample multiplexing and deduplication for accurate variant calling. In the final step, the amplicons are pooled and purified prior to sequencing.
